8-Propyl-6H-[1,3]dioxolo[4,5-g]chromen-6-one: A new coumarin with monoamine oxidase B inhibitory activity and possible anti-parkinsonian effects BJPS
Olaya,María del Pilar; Vergel,Nadezdha Esperanza; López,Jose Luis; Viña,Dolores; Guerrero,Mario Francisco.
Parkinson’s disease is a common neurodegenerative disorder. In this study, the monoamine oxidase inhibitory activity and potential anti-parkinsonian effects of 8-propyl-6H-[1,3]dioxolo[4,5-g]chromen-6-one (FCS303), a new synthetic coumarin, were evaluated. To do this, we used the reserpine model of Parkinson’s disease, an assay of levodopa/carbidopa potentiation, the catalepsy model of haloperidol, and an in vitro assay against monoamine oxidase (MAO) activity. Additionally, lipid peroxidation and protein carbonyl group quantification was performed in mice brain homogenates previously treated with haloperidol. FCS303 inhibited monoamine oxidase B (MAO-B) with an IC50 of 5.46 ± 0.36 µM; however, there was no effect on monoamine oxidase A (MAO-A). The oral...

A critical evaluation of the effect of population size and phenotypic measurement on QTL detection and localization using a large F2 murine mapping population Genet. Mol. Biol.
Li,Xinmin; Quigg,Richard J; Zhou,Jian; Xu,Shizhong; Masinde,Godfred; Mohan,Subburaman; Baylink,David J..
Population size and phenotypic measurement are two key factors determining the detection power of quantitative trait loci (QTL) mapping. We evaluated how these two controllable factors quantitatively affect the detection of QTL and their localization using a large F2 murine mapping population and found that three main points emerged from this study. One finding was that the sensitivity of QTL detection significantly decreased as the population size decreased. The decrease in the percentage logarithm of the odd score (LOD score, which is a statistical measure of the likelihood of two loci being lied near each other on a chromosome) can be estimated using the formula 1 - n/N, where n is the smaller and N the larger population size. This empirical formula has...

Acute buspirone abolishes the expression of behavioral dopaminergic supersensitivity in mice BJMBR
Queiroz,C.M.T.; Alcântara,F.B.; Yagüe,A.M.L.; Bibancos,T.; Frussa-Filho,R..
Previous studies have shown that rats withdrawn from long-term treatment with dopamine receptor blockers exhibit dopaminergic supersensitivity, which can be behaviorally evaluated by enhanced general activity observed in an open-field. Recently, it has been reported that co-treatment with the non-benzodiazepine anxiolytic buspirone attenuates the development of haloperidol-induced dopaminergic supersensitivity measured by open-field behavior of rats. The aims of the present study were: 1) to determine, as previously reported for rats, if mice withdrawn from long-term neuroleptic treatment would also develop dopaminergic supersensitivity using open-field behavior as an experimental paradigm, and 2) to examine if acute buspirone administration would...

After restraint stress, mice with high and low immobility time have a different neurochemical and behavioral profile MV&Z
Reis-Silva, Thiago; Lima, Ana Paula; Moreira, Natalia; Calefi, Atilio; Sandini, Thaísa Meira; Florio, Jorge Camilo; Bernardi, Maria Martha.
High and low immobility on the tail suspension test is a common tool for assessing antidepressant drug effects. However, those parameters might be a natural phenotype present in the mice population and could be selected through behavior evaluation without an induced-drug behavior need. High immobility has been also related to depressed-like behavior, that in humans are characterized by wide range of symptoms, such as anhedonia, anxiety and tiredness.

Aggravation of pathogenesis mediated by aflatoxin B1 in mice infected with Trypanosoma brucei rhodesiense OAK
Kibugu, J.K; Makumi, J.N; Ngeranwa, J.J.N; Kagira, J.M; Gathumbi, J.K; Mwangi, J.N.
Aflatoxins are known to alter the pathogenesis of many infectious diseases, but such effects have not been evaluated in trypanosome infections. The aim of the present work was to assess the effects of aflatoxin B1 on the pathogenesis of Trypanosoma brucei rhodesiense infection using a murine model. Mice fed on 0.50 mg/kg aflatoxin b. wt. were infected with T. b. rhodesiense and compared to trypanosome infected and uninfected aflatoxin-fed controls. The clinical and pathological changes were determined and the quantitative data statistically analysed using standard methods. The results showed that infected aflatoxin-fed mice had pronounced dyspnoea, significantly (P<0.05) reduced survival, extreme emaciation, pronounced macrocytic normochromic anaemia...

Alanyl-glutamine attenuates 5-fluorouracil-induced intestinal mucositis in apolipoprotein E-deficient mice BJMBR
Araújo,C.V.; Lazzarotto,C.R.; Aquino,C.C.; Figueiredo,I.L.; Costa,T.B.; de Oliveira Alves,L.A.; Ribeiro,R.A.; Bertolini,L.R.; Lima,A.A.M.; Brito,G.A.C.; Oriá,R.B..
Apolipoprotein E (APOE=gene, apoE=protein) is a known factor regulating the inflammatory response that may have regenerative effects during tissue recovery from injury. We investigated whether apoE deficiency reduces the healing effect of alanyl-glutamine (Ala-Gln) treatment, a recognized gut-trophic nutrient, during tissue recovery after 5-FU-induced intestinal mucositis. APOE-knockout (APOE-/-) and wild-type (APOE+/+) C57BL6J male and female mice (N=86) were given either Ala-Gln (100 mM) or phosphate buffered saline (PBS) by gavage 3 days before and 5 days after a 5-fluorouracil (5-FU) challenge (450 mg/kg, via intraperitoneal injection). Mouse body weight was monitored daily. The 5-FU cytotoxic effect was evaluated by leukometry. Intestinal villus...

Amylin induces hypoglycemia in mice Anais da ABC (AABC)
Guerreiro,Luiz H.; Silva,Daniel DA; Sola-Penna,Mauro; Mizurini,Daniella M.; Lima,Luís M.T.R..
Amylin is a 37-aminoacid pancreatic protein that exerts control over several metabolic events such as glycemia and lacticemia. Amylin has long been shown to induce increases in arterial plasma glucose. We decided to investigate whether amylin plays additional roles in the glucose metabolism. We evaluated glucose homeostasis using whole blood from the tail tip of fasting, conscious, unrestrained normal and streptozotocyn-induced diabetic mice following subcutaneous administration of mouse amylin. Subcutaneous injection of 1 μg mouse amylin caused a transient decrease in whole blood glucose in both normal and diabetic mice in the absence of insulin. The blood glucose levels were lowest approximately 2 hours after amylin administration, after that they...

Salmonella enterica serovar Enteritidis is one of the main pathogens responsible for foodborne illness in Brazil. Probiotic bacteria can play a role in defense and recovery from enteropathogenic -infections. In this study, the ability of Lactobacillus acidophilus LA10 to colonise and exert anta-gonistic effects in the gastrointestinal tract was tested before and during experimental infection in conventional mice contaminated with S. Enteritidis (SE86). A dose of 0.1 mL containing 10(8) viable cells of SE86 and L. acidophilus LA10 was orally administered by gavage to mice. The experiment was divided into groups. As a negative control, Group 1 was administered only sterile saline solution. As a positive control, Group 2 was administered only SE86. Group 3...

Antioxidant capacity of vitamin C in mouse liver and kidney tissues Biocell
Weyers,Alicia; Ugnia,Laura I.; García Ovando,Hugo; Gorla,Nora B..
In the present study, the antioxidant capacity of vitamin C was examined in the liver and the kidney tissues of mice with or without ciprofloxacin (CFX) treatment. The antioxidant capacity of the vitamin was evaluated in terms of lipid hydroperoxides (LOOH) and thiobarbituric acid reactive substances (TBARs). The experimental design was 15 days of water (control and CFX groups) or vitamin C (vitamin C and vitamin C plus CFX groups) in drinking water. One dose of CFX was injected, 15 minutes before sacrifice, in the corresponding mice. The initial nmol of lipid hydroperoxides/g of tissue were 137 ± 11 in the kidney and 145 ± 15 in the liver, and the nmol of TBARs were 13 ± 0.7 and 12 ± 0.6, respectively. Pre-treatment with vitamin C reduced the levels of...

Assessment of the cytotoxic, genotoxic, and antigenotoxic activities ofCeltis iguanaea (Jacq.) in mice Anais da ABC (AABC)
BORGES,FLAVIO F.V.; MACHADO,THIAGO C.; CUNHA,KÊNYA S.; PEREIRA,KARLA C.; COSTA,ELSON A.; PAULA,JOSÉ R. DE; CHEN-CHEN,LEE.
Ethnobotanical surveys of Cerrado native plants show that leaves of Celtis iguanaea (Jacq.) Sargent (Cannabaceae), popularly known in Brazil as “esporão de galo”, are used in folk medicine for body pain, asthma, cramps, poor digestion, urinary infection, kidney dysfunctions, as well as a stimulant and diuretic. This work aimed at evaluating possible C. iguanaea aqueous leaf extract (CALE) cytotoxicity, genotoxicity, and antigenotoxicity using the mouse bone marrow micronucleous test. To assess CALE genotoxicity, Swiss mice were orally treated with three different extract concentrations (100, 300, and 500 mgkg−1). To evaluate its antigenotoxicity, the same doses were used simultaneously with a single i.p. dose of mitomycin C (MMC, 4mg.kg−1). The frequencies...

Behavioral changes induced by cocaine in mice are modified by a hyperlipidic diet or recombinant leptin BJMBR
Erhardt,E.; Zibetti,L.C.E.; Godinho,J.M.; Bacchieri,B.; Barros,H.M.T..
The objective of the present study was to determine if the acute behavioral effects of cocaine acutely administered intraperitoneally (ip) at doses of 5, 10 and 20 mg/kg on white male CF1 mice, 90 days of age, would be influenced by leptin acutely administered ip (at doses of 5, 10 and 20 µg/kg) or by endogenous leptin production enhanced by a high-fat diet. The acute behavioral effects of cocaine were evaluated in open-field, elevated plus-maze and forced swimming tests. Results were compared between a group of 80 mice consuming a balanced diet and a high-fat diet, and a group of 80 mice fed a commercially available rodent chow formula (Ralston Purina) but receiving recombinant leptin (rLeptin) or saline ip. Both the high-fat-fed and rLeptin-treated mice...

Beneficial Effect of Hyperbaric Oxygen Therapy on the Follicular Survival in the Mouse Ovary after Transplantation OAK
HAYASHI, Satomi; ASANO, Tomoyoshi; KAKIZAKI, Ryujiro; SUZUKI, Hiroshi; 鈴木, 宏志.
A large proportion of follicles are lost during the initial ischemia that occurs after transplantation of ovarian tissues. Thus, the effect of hyperbaric oxygen therapy (HBO) on the follicular loss of ovarian tissues after transplantation was examined in mice. Ovarian slices from ICR mice were transplanted under the kidney capsule in ovariectomized ICR. Hyperbaric oxygen with 100% oxygen was initiated for 30 min at 2.5 atmospheres absolute immediately after transplantation, and this treatment was repeated at 48-h intervals for 2 weeks. The number of follicles was dramatically reduced at 2 weeks post transplantation. However, HBO was significantly effective in enhancing the survival of transplanted ovarian follicles. The survival rates of primordial and...

Bovine respiratory syncytial virus: immunohistochemichal detection in mouse and bovine tissues using a Mab against human respiratory syncytial virus Arq. Bras. Med. Vet. Zootec.
Almeida,R.S.; Spilki,F.R.; Roehe,P.M.; Verinaud,L.M.C.; Arns,C.W..
An immunoistochemical (IHC) test was developed to detect bovine respiratory syncytial virus (BRSV) in cell cultures and tissues of experimentally infected mice and calves, using a commercial monoclonal antibody (Mab) against human respiratory syncytial virus (HRSV), as a less expensive alternative, instead of producing specific monoclonal antibodies to BRSV. Clinical samples from calves suffering respiratory disease were also submitted to this test. IHC detected BRSV antigens in mouse tracheas (3, 5 and 7 days post-infection) and lungs (5 and 7 days post-infection), and in one of three lungs from experimentally infected calves. Lungs samples from two naturally infected calves were tested and resulted positive for BRSV by the IHC test. These results suggest...

Cell-mediated immune response to Leishmania chagasi experimental infection of BALB/c immunosuppressed mice J. Venom. Anim. Toxins incl. Trop. Dis.
Machado,JG; Hoffmann,JL; Krause,VLK; da Silva,AV; Dias-Melicio,LA; Langoni,H.
Leishmaniasis, a zoonosis of worldwide distribution, presents a significant impact on immunosupressed patients. This study aimed to evaluate Leishmania chagasi infection in BALB/c mice immunosuppressed with dexamethasone. Spleen cells stimulated or not with L. chagasi were cultured for cytokine quantification (IFN-γ, IL-2, IL-4 and IL-10) by sandwich ELISA. Parasite loads in the spleen and liver were determined by means of culture microtitration. Immunosuppressed groups showed statistically lower spleen weight and CD4-cell percentage in blood on the day of infection and produced Th1 and Th2 cytokines on other days of the study. The other infected groups, weather immunosupressed or not, also produced Th1 and Th2 cytokines. Parasite loads in the spleen and...

Co-culture of Mouse Two-cell Embryos on Human Endometrial Stromal Cells in Proliferative and Secretory Phases International Journal of Morphology
Fakheri,Taravat; Nankali,Anisodowleh; Khazaei,Mozafar; Ghanbari,Ali; Abedini,Mehrandokht; Saeidiborojeni,Hamidreza.
There were no significant differences in the distribution of embryos reaching to 2- cells, 4- cells, morula or blastocysts culturing on human endometrial stromal cells (Secretory or proliferative phases). The percent of morula in stage A (without fragmentation), stage B (<25% fragmentation), stage C (25-50% fragmentation) and stage D (>50% fragmentation) and did not showed significant differences between two coculture groups. Thus, the phase that the endometrial stromal cells were in thereby did not affect on the quality of embryos.

Combined effects of diethylpropion and alcohol on locomotor activity of mice: participation of the dopaminergic and opioid systems BJMBR
Gevaerd,M.S.; Sultowski,E.T.; Takahashi,R.N..
The widespread consumption of anorectics and combined anorectic + alcohol misuse are problems in Brazil. In order to better understand the interactive effects of ethanol (EtOH) and diethylpropion (DEP) we examined the locomotion-activating effects of these drugs given alone or in combination in mice. We also determined whether this response was affected by dopamine (DA) or opioid receptor antagonists. A total of 160 male Swiss mice weighing approximately 30 g were divided into groups of 8 animals per group. The animals were treated daily for 7 consecutive days with combined EtOH + DEP (1.2 g/kg and 5.0 mg/kg, ip), EtOH (1.2 g/kg, ip), DEP (5.0 mg/kg, ip) or the control solution coadministered with the DA antagonist haloperidol (HAL, 0.075 mg/kg, ip), the...

Combined oral toxicity of azaspiracid-1 and yessotoxin in female NMRI mice ArchiMer
Aasen, John A. B.; Espenes, Arild; Miles, Christopher O.; Samdal, Ingunn A.; Hess, Philipp; Aune, Tore.
For many years, the presence of yessotoxins (YTXs) in shellfish has contributed to the outcome of the traditional mouse bioassay and has on many occasions caused closure of shellfisheries. Since YTXs do not appear to cause diarrhoea in man and exert low oral toxicity in animal experiments, it has been suggested that they should be removed from regulation. Before doing so, it is important to determine whether the oral toxicity of YTXs is enhanced when present together with shellfish toxins known to cause damage to the gastrointestinal tract. Consequently, mice were given high doses of YTX, at 1 or 5 mg/kg body weight, either alone or together with azaspiracid-1 (AZA1) at 200 μg/kg. The latter has been shown to induce damage to the small intestine at this...
